The only exceptions are medbays and science labs, where the production per tile is most efficient at 2 wide. "Almost all rooms are more efficient at 3 wide compared to 1 or 2 wide. I have designed my Vault using the same basic layout as seen in this video Some are very organized and built strategically, placing power rooms at the top, where others have been completely random builds where the layout makes no sense, rooms placed just "wherever" throughout the vault. I have seen dozens of videos which claim to be "perfect vault" builds. I like to do plenty of research via you-tube and discussions in order to maximize my time and effort in a game, rather than alot of trial and error and rebuilding. I am very new to this game so I am still in the "learning" stage.
